The eight teams who finished as runners-up in the group stage were paired with the eight clubs transferred into the competition from the Champions League as follows.
Play-off round
- Feyenoord v AS Roma
- AC Milan v Rennes
- Lens v Freiburg
- Young Boys v Sporting CP
- Benfica v Toulouse
- Braga v Qarabag
- Galatasaray v Sparta Prague
- Shakhtar Donetsk v Marseille
These fixtures will be played over two legs in February.
Liverpool automatically progressed to the Round of 16 after topping Group E and will therefore face one of the eight sides who advance from the play-off round.
